5.5 Zusammenfassende Wertung
Die ausgewerteten in-vitro-Untersuchungen haben gezeigt, daß die Protozoenzahl im RUSITEC stabil ist, die angewandten Zählmethoden mit der modifizierten Nageotte-Zählkammer zuverlässig sind und die Protozoenzahlen empfindlich auf fütterungsbedingte Veränderungen reagierten [mit Epicoccum nigrum, Alternaria alternata, Mucor racemosus oder Ulocladium chartarum verschimmeltes Heu, chronische Pansenazidosen mit und ohne Thiaminzulagen, Schwefelsupplementierungen und Thiamingaben (allein)].
Im einzelnen konnten folgende Auswirkungen beobachtet werden:
- Rückgang der mittelgroßen Protozoen durch Epicoccum (-27 %), Alternaria (-16 %) und Mucor (-9 %)
- Geringgradiger Anstieg der mittelgroßen Protozoen durch Ulocladium (+2 %)
- Erhaltung einer Protozoenpopulation während chronischer Pansenazidose bei Thiaminsupplementierung im Gegensatz zu Versuchen unter Thiaminmangel
- Anstieg der kleinen Protozoen (+30 %) nach Zulage von 0,125 mg Sulfit/ml Pansensaft gegenüber einem Rückgang der kleinen um 55 % und der mittelgroßen um bis zu 100 % nach Gabe von 0,5 mg Sulfat/ml Pansensaft
- Allgemeine Stabilisierung der kleinen Protozoen durch Thiamin bei gleichzeitiger Erhöhung der Schadwirkung von Sulfit (-10 %) und Schadgras (-11 %) auf selbige Protozoenfraktion
- Erhöhung der Schadwirkung von Sulfit (-17 %) auf mittelgroße Protozoen durch Thiamin
- Kompensierung negativer Schadgraseinflüsse auf die mittelgroßen Protozoen durch Thiamin um +10 %
- Förderung des positiven Ulocladiumeinflusses durch Thiamin um +26 %
Hierbei sind direkte und indirekte Futtermitteleinwirkungen auf die Protozoen zu vermuten.
Genaue Mechanismen sind im einzelnen jedoch noch nicht abgeklärt und bedürfen weiterer Untersuchungen.
Allgemein betrachtet reagieren Protozoen auf intraruminale Milieuveränderungen, so daß deren Anzahl und Konzentrationsänderungen als Gesundheitsindikatoren bei Krankheitsverdacht oder zur Abschätzung des Schweregrads einer Erkrankung dienen können (z. B. bei subakut bis chronischen Pansenazidosen, bei Schwefelintoxikationen und Mykotoxin-Vergiftungen). So werden Einzeller auch in der Literatur bereits vielfach als Biosensoren für „bioassays“ in in-vitro-Versuchen eingesetzt (DIVE et al. 1978; NISTIAR et al. 1986; BIJL et al. 1988; NISHIE et al. 1989; BENITEZ et al. 1994).
Tierindividuelle, diurnale und saisonale Schwankungen der Protozoenkonzentrationen müssen jedoch bei der Beurteilung des Pansensaftes ebenso bedacht werden wie die große Spannbreite der Protozoenkonzentration. Deshalb sollte zur Bewertung der Protozoenanzahl entweder deren Konzentrationsverlauf betrachtet oder aber Pansensaft gesunder Tiere der gleichen Herde als Vergleich herangezogen werden.
6. Zusammenfassung
In jeweils 25 Tage dauernden in-vitro-Versuchen (RUSITEC) wurden in praxi bedeutsame Einflüsse auf das Pansenmilieu des Rindes simuliert.
Diese Einflüsse waren:
- Grundfutter (Gramineae), verpilzt mit a) Epicoccum nigrum
b) Alternaria alternata c) Mucor racemosus d) Ulocladium chartarum
- Schadgras (natürlich grobsinnlich verschimmelt) - Sulfit/Sulfat
- Chronische Pansenazidose
Gleichzeitig wurde derart belastetem Pansenmilieu Thiamin zugelegt. Ziel dieser Untersuchungen war die Überprüfung der Wirkung oben aufgeführter Bedingungen auf die Konzentration der Protozoen im Pansensaft.
Zur Zählung und Differenzierung der Protozoen (kleine, mittelgroße und große) wurden zwei Zählmethoden unter Verwendung einer modifizierten Nageotte-Zählkammer miteinander verglichen und als geeignet und zuverlässig erkannt (VK für die Summe aller Protozoen 5 - 5,6 %, VK für die Anzahl kleiner Protozoen 5,9 – 6,0 %, VK für die Anzahl mittelgroßer Protozoen 12,7 – 13,6, % VK für die Anzahl großer Protozoen 22,3 – 25,5 %).
Die Ergebnisse sind in Tab. 6.1 zusammengefaßt.

7. Summary
The influence of mouldy grass, moulded hay (kind of moulds: Epicoccum nigrum, Alternaria alternata, Mucor racemosus, Ulocladium chartarum), acidotic conditions, sulphur and the efficiency of thiamine substitution on rumen protozoa was investigated using the longterm rumen simulation technique (RUSITEC) for about 25 days.
Two different counting methods for the total number of protozoa and their differentiation into small, medium-sized and large seizes were compared using a modified Nageotte-counting cell. They were recognized as suitable and reliable (VK for the total number of protozoa 5 – 5.6 %, VK for the number of small protozoa 5.9 – 6.0 %, VK for the number of medium-sized protozoa 12.7 – 13.6 %, VK for the number of large animals 22.3 – 25.5 %).
The following effects of the above mentioned agents with and without thiamine addition on rumen protozoa could be noted:
