Autosomal dominanter Erbgang
Ein normales ("Wildtyp") und ein betroffenes ("Mutation") Allel
Aktivierende Mutationen (RET)
Wenige spezifische Mutationen in konservierten Genabschnitten
Inaktivierende Mutationen (alle anderen)
Sehr heterogene Arten von Mutationen
Über das ganze Gen verstreut

Familiar versus "apparently sporadic" = clinical versus genetic identification of
heritable risk
Family history becomes positiv in at least 60% of index patients
Difficulty to elicit a potentially heritable familial background
Incidents of unexplained deaths, cardiac arrests, stroke and hypertension are to common and to unspecific
asking for the occurrence of pheochromoytoma and paragangliomas will probably remain inconclusive
important issue is to raise the awareness of the possibility of a familial background
will eventually lead to a better evaluation of the familial history and to decrease the number of "apparently sporadic" cases
