Study on bronchoconstriction induced by histamine, serotonin or passive sensitization in “Precision Cut Lung Slices“ of the horse
The aim of this study was to examine the effects of histamine, serotonin, their inhibitors, and the effect of the H4-agonist 4-methylhistamine on equine bronchoconstriction with precision-cut lung slices (PCLS), a technique established by VIETMEIER (2004). Additionally we investigated if the model of passive sensitization established in the rat by WOHLSEN et al. (2001) can be applied to the horse.
14 horses were available for this study. A general clinical examination and an examination of the respiratory tract (including arterial blood gas analysis and bronchoscopy followed by analysis of aspirated tracheobronchial secretions) was performed one day before euthanasia. The results obtained were used to divide the horses in groups according to evidence of pulmonary disease.
Immediately post mortem the Lobus accessorius was removed through an intercostal approach and filled with 1,5 % agarose solution. PCLS were obtained from the lung tissue using a microtome. Vitality was verified before and after every contraction series with a methacholine-induced (10-5 mol/l) bronchoconstriction. In the first contraction series the concentration of histamine and serotonin was increased after each application on the PCLS, from 10-7 to 10-4 mol/l for histamine and 10-8 to 10-5 mol/l for serotonin. In the second contraction series the effects of histamine receptor antagonists (cetirizin, rantidin, thioperamid, JNJ7777120) and a serotonin receptor antagonist (ketanserin) were tested. This was done by incubating the PCLS with one particular receptor antagonist for 30 minutes. The contraction status of the bronchi was photographed in defined intervals and digitally evaluated using two image
analysis. With this method it was possible to evaluate the extent of bronchoconstriction in comparison to the initial lumen.
Histamine caused a concentration-dependent bronchoconstriction, which was slightly significant (p=0,05) starting at a concentration of 10-5 mol/l. The histamine induced bronchoconstriction could be inhibited by a previous incubation period with the H1 -receptor antagonist cetirizin. However, prior incubation with the antagonists ranitidin, thioperamid and JNJ7777120 showed no significant effects.
Serotonin showed a concentration-dependent bronchoconstriction in one of the six horses, which could be inhibited by prior incubation with ketanserin.
4-methylhistamine did not induce bronchoconstriction in any of the vital PCLS.
For the passive sensitization the PCLS were incubated in serum from a horse actively sensitized against ovalbumin. As control group serum from a horse with no detectable antibodies against this allergen was used. The response of the bronchi was photographed two and ten minutes after the addition of ovalbumin. While two horses showed no reaction, a considerable constriction could be detected in the PCLS of seven horses in both groups. One horse showed a significant reaction only in the passively sensitized slices.
Due to the high number of slices that can be obtained from one lung, the PCLS-technique allows a reduction of the number of experimental animals. In the present study we were able to gain an insight into the basic effects that histamine and serotonin have on the bronchi of horses and the ability of specific receptor antagonists to prevent these effects. There were no clear findings for the passive sensitization of PCLS of the horse. For further studies on a model of passive sensitization an allergen should be used to which the horses have not come into contact under normal conditions.
