2.5.2 Färbeprotokolle
In den Färbeprotokollen ist jeweils nur ein Beispiel für eine Einfach-, eine Doppel- und eine Dreifachfärbung ausführlich dargestellt (in der Übersichtstabelle Tab. 2.6 mit Sternchen* markiert). Der nachfolgenden Tabelle sind die Änderungen für die ent-sprechende Färbung zu entnehmen und im Protokoll an der erforderlichen Stelle aus-zutauschen. Die Färbeprotokolle für die Rekonstruktion des PDF-Netzwerks in Ganz-hirnen (einmal mit Luzifer Yellow, einmal mit anti-Synapsin) sind vollständig aufgeführt.

Färbeprotokoll für Einzelfärbung anti-PDH
1. Präparation • Bienenköpfe in Paraffin befestigen
• in Phosphatpuffer (PB, pH 7,4) Gehirne herauspräparieren
• in Blockschälchen mit PB Tracheen entfernen
2. Fixierung • in Schnappdeckelgläschen mit Zamboni’s Fixativ
mindestens 24 Std.
bei 4°C
3. Einbetten • in Gelatine-Albumin 42°C, max. 50°C 4. Fixierung • in Schnappdeckelgläschen
mit 4 % Paraformaldehyd (PFA)

5. Vibratom • in PB schneiden
• Sektionen überführen in Mikrotiterplatte mit PB

6. Waschen • 3 x mit PB
