antigen

LI-CAMis acell adhesion molecule thatplays
animportant
role inthe
development
offetal brain. Mutations in the LI-CAMgene areconnectedwithheavy neurological pathologies,
thataresummarizedunder the nameofCRASHSyndrome.
The
protein,
which waspurified by
means ofmAbchCE7,
showed a deletion of five amino acidsattheN-Terminus.corresponding
to a deletion ofexon2 of Ll-CAMgene.Partial
cloning by
RT-PCR withLI-specific primers
showed that in neuroblastoma cells bothisoforms,
the„füll-length"
and aexon2'exon27-deletedformwerepresent.
Additionalrenal Carcinoma cells weretested for the
expression
of Ll-CAM: five of sixwere found
positive
andexpressed only
the deleted isoform.Only
aweak reaction wasfound when normaladult renal tissue was
analyzed,
in contrast normalhuman braincortex
tissue,
which reactedstrongly
with chCFJ.Additionally,
fifteen renaltumortissueslices of different
origin
weretested: five ofthemwerepositive.
Northern blot
analyses
showed that in the tumor cells a correlation exists betweenprotein-
and mRNAexpression.
As well. Ll-CAMmRNAwas found in normaltissues likebrain, kidney,
smallintestine,
stomach andplaccnta.
In contrast,Ll-CAM mRNA couldnotbe found in heart. liver,colon, hing, muscle, spieen

Das
Antigen
wurde durchImmunafflnitätschromatographie
aus Zellextrakten von 2x108 Zellen(SK-N-BE(2)c
undFöhn) gereinigt. Antigcn-positive
Fraktionen wurdenimmunpräzipitiert
und mit SDS-PAGE unter reduzierendenBedingungen aufgetrennt.
Die
Bande,
die demAntigen entsprach,
wurde aus WesternBlots(PVDF-Membran) ausgeschnitten und,
nachDeblockierung
desN-Terminus,
wurden fünfzehn bzw.achtzehn
Aminosäuren (SK-N-BE(2)c respektiv Föhn)
des N-terminalen Teiles des Proteinssequenziert.

Protein. Ll-CAM ist eineZelladhäsionsmolekül,
das einewichtige
Rolle bei der
Entwicklung
vonfoetalemGehirnspielt.
Mutationen im Ll-CAM Gensind mit schwerenneurologischen Pathologien verbunden,
die unter dem Namen CRASII-Syndrom zusammengefasst
werden.Das
Protein,
welches mittels mAkch.CE7gereinigt wurde,
wargekennzeichnet
durcheine DeletionvonfünfAminosäurenam
N-Terminus,
die auf dem LI -Gen einer DeletionvomExon 2
entsprach.
PartielleKlonierung
durch RT-.PCR mitLI-spezifischen
Primerszeigte,
dassinNeuroblastomazellendie beidenIsoformen,
die„fii.Il-l.en.ght"
Form und dieForm mit einer Deletion sowohl im Exon 2 als auch im Exon 27 vorhandenwaren.
Weitere Nierenkarzinomzellen wurden auch auf die
Expression
von Ll-CAMgetestet:
fünfvon sechs waren
positiv
und enthalten nur die deletierte Isoform. Nur eine seh¬schwache Reaktion konnte
nachgewiesen
werden wenn normales erwachsenesNierengewebeanalysiert
wurde,hingegen
eine starke Reaktion mit dem normalenHirn- Cortex Gewebe wurdenachgewiesen.
Zusätzlich wurden auch fünfzehnNierentumorgewebe
verschiedenenUrsprungs getestet:
fünfdavonwarenpositiv.

Analysen zeigten,
dasses in den Tumorzetlen eine Korrelationzwischen Protein- undmRNA-Expression
existiert. Ll-CAM mRNA wurde ebenso in normale Geweben wieHirn, Nieren, Dünndarm, Magen
und Plazentagefunden. Flingegen,
inHerz, Leber,
Kolon.Milz, Lungen.
Muskel und Lloden konnte kern Ll-CAM mRNAnachgewiesen
werden.Ein Teil der Nierentumoren
zeigt genetische
Aberrationenin ChromosomXq28,
wo dasLl-CAM Gen lokalisiert ist.
Analysen
von weiterenNierentumorgeweben
sindvorgesehen,
um einemögliche
Korrelation zwischengenetischen Veränderungen
indiesen Tumorenund

unimportante
ruolo nellosviluppo
del cervello del feto. Mutazioni del gene della Ll-CAM provocano
gravi patologie neurologiche
che sonoriassuntesotto il nomedisindrome di CRASH.La
proteina, purificata
tramiteFanticorpo chCJE7,
mostra una delezione dicinque
aminoacidi nella
regione
delN-terminus, questa corrispon.de
ad una delezione delsecondo exonedel gene della Ll-CAM. Laclonanzione
parziale
del gene dellaLl-CAM,
tramite RT-PCR con
primers specifici.
ha mostrato la presenza, nelle cellule del neuroblastoma di entrambe le isoforme dellaLl-CAM,
cioe sia la formaoriginale
chequella
condelezioni del secondo e delventisettesimo exone.Le cellule di sei divarsi
tipi
diCarcinomarenale sono State esaminatesull'espressione
dellaLl-CAM.
Cinque esprimono
laLl-CAM,
ma soltanto l'isoforma con entrambe le delezioni. Una debole reazione confanticorpo
e stata rilevata all'analisi di normaletessuto renale umano,mentre cot normale tessuto cerebrale umano la reazionee stata
molto
piü
marcata.Altriquindici
tessuti tumorali renali diorigine
diversa sono stallesaminati:unterzoha
reagito
confanticorpo.

Riassunto Le analisi tramite il Northern, blot hanno rilevato
che,
nelle celluletumorali,
esiste unacorrelazione tra
l'espressione
dellaproteina
equella
del RNA messaggero. II mRNAdella Ll-CAMepresente
nei tessuti normaliumani delccrvcllo,
rene, intestinotenue,
stomacoe
placenta.
Per contro. non ce n'era traccia nei tessuti dei cuore,fegato, colon, milza, polmone,
muscoloetesticoli.Alcuni tumori renali
presentano
aberrazionigenetiche
sul cromosomaXq28,
dove elocalizzato il gene della Ll-CAM. L'analisi di ulteriori tessuti tumorali renali sara
necessaria per
poter
individuareunapossibile
correlazione tra le modificazionigenetiche
in
questi
tumorie le variazionidelfespressione
dellaLl-CAM. AIfine dipoter prendere
in cosiderazione un'eventuale
applicazione deU'anticorpo
chCE7 nelladiagnostica
e nellaterapia
di alcuni tumorirenali,
cosi come e stato fatto per ilneuroblastoma,
ulteriorianalisi saranno necessarie. Una
mighore
caratterizzazione bioclfimica delle diverse isoforme della Ll-CAM fornirebbe ulterioriiirformazioni,
e, inparticolare,
sequeste
proteine
sono caratterizzatedaspecific!
determinanti tumorali e comela loroespressione
e
regolata
nelle cellule tumorali.11
