Ziel dieser Dissertation war es, grundlegende Kenntnisse über den SMP des porcinen Colons, besonders in Hinblick auf seine Mechanosensitivität, zu erhalten.
Aufgrund der Ergebnisse der vorliegenden Arbeit kann bestätigt werden, dass eine Subpopulation enterischer Nervenzellen des ISP sowohl im Colon ascendens als auch im Colon transversum mechanisch wirkende Kräfte (Kompression und Dehnung) wahrnimmt und mit neuronaler Aktivität reagiert.
Im ENS von Meerschweinchen wird bereits von der Existenz multimodaler Neurone, die sich sowohl als kompressions- als auch als dehnungssensitiv erweisen, ausgegangen (MAZZUOLI-WEBER u. SCHEMANN 2015a). Mittels eines gepaarten Versuchsaufbaus gilt es herauszufinden, ob solche multimodalen Neurone auch im porcinen ENS existieren. Weiterhin sollten auch andere porcine Darmabschnitte hinsichtlich einer möglichen Existenz von MEN in zukünftigen Arbeiten untersucht werden.
Untersuchungen hinsichtlich der funktionellen Einordnung der hier identifizierten MEN könnten ebenfalls künftig in Erwägung gezogen werden. Die retrograde Tracing-Technik ist beispielsweise in der Lage, Interneurone, Motoneurone oder sensorische Neurone zu markieren und somit zu identifizieren.
Weitere Experimente sind in Zukunft außerdem notwendig, um zu klären, welche Ionenkanäle mechanische Signale im SMP des Schweins kodieren und elektrische Antworten vermitteln. KREUTZ (2019) konnte im Colon descendens von Meerschweinchen eine wichtige Rolle von Kationenkanälen nachweisen, indem er Experimente mit dem unspezifischen Blocker mechanisch gesteuerter Kationenkanäle Gandolinium (III)-Chlorid-Hexahydrat durchführte.
Die vorliegende Arbeit bietet erste wichtige Einblicke in die neurochemische Codierung porciner MEN. Zukünftig sollten zusätzliche Neurotransmitter, wie beispielsweise der Transmitter inhibitorischer Synapsen VIP, hinsichtlich ihrer Häufigkeit und Verteilung analysiert werden, um weitere Transmitter-Kombinationen der MEN aufzudecken und

Charakterisierung mechanosensitiver Neurone im submukösen Plexus des porcinen Colons
Die Neurone des enterischen Nervensystems (ENS), das mit seinen beiden Plexus direkt in der Wand des Magen-Darm-Traktes lokalisiert ist, sind permanent der Aktivität von Zirkulär- und Longitudinalmuskulatur ausgesetzt. Neben den Neuronen des myenterischen Plexus (MP), der sich zwischen den beiden intestinalen Muskelschichten befindet, ist bekannt, dass auch die Neurone des in der Submukosa lokalisierten submukösen Plexus (SMP) direkt durch mechanische Stimuli aktiviert werden. Daten zur Mechanosensitivität porciner submuköser Neurone fehlen bislang, wenngleich das Schwein, basierend auf strukturellen und physiologischen Gemeinsamkeiten des Gastrointestinaltraktes, als relevantes Modelltier für den Menschen diskutiert und genutzt wird.
Ziel der vorliegenden Arbeit war es, grundlegende Kenntnisse über die Neurone des SMP im porcinen Colon, insbesondere in Bezug auf ihre Mechanosensitivität, zu erlangen. Dabei wurde zunächst ein Protokoll etabliert, welches es ermöglichte, neuronale Signale in Lebendpräparaten des porcinen Colon ascendens und transversum optisch darzustellen. Als bildgebendes Verfahren wurde die Ultrafast-Neuroimaging-Technik in Verbindung mit dem spannungssensitiven Farbstoff Di-8-ANEPPS verwendet, sodass auf Einzelzellebene alle Neurone eines Ganglions gleichzeitig untersucht werden konnten. Da die Ganglien des SMP beider untersuchter Darmlokalisationen bereits im Durchlicht des Lichtmikroskops anhand ihrer Größe, Morphologie und Oberflächenstruktur eindeutig identifiziert werden konnten, konnte auf eine Vorfärbung der Neurone verzichtet werden. Die Vitalität ausgewählter Ganglien wurde zunächst überprüft, indem eine elektrische (mittels Punktstimulationselektrode) oder chemische Stimulation (mit dem nikotinergen Acetylcholin-Rezeptor Agonisten Nikotin) der Neurone erfolgte. Mit beiden Verfahren war es möglich, Neurone der untersuchten Colonabschnitte zu aktivieren. Einen

wesentlichen Bestandteil der vorliegenden Arbeit stellten mechanische Stimulationen dar. Dazu zählten sowohl Kompressionen mittels intraganglionärer Krebslösung-Injektionen als auch Dehnungen submuköser Neurone. Ein eigens entwickeltes Stretching Tool erlaubte erstmals die bidirektionale Dehnung gesamter Ganglien in entgegengesetzte Richtungen. Beide Stimuli hatten reproduzierbare elektrische Entladungen in einer Subpopulation enterischer Nervenzellen zur Folge, wobei weder alters- noch geschlechtsbedingte Effekte festgestellt werden konnten. Die immunhistochemische Untersuchung der mechanosensitiven enterischen Neurone (MEN) bezüglich ihrer Cholinacetyltransferase (ChAT)-, Substanz P (SP)- und Stickstoffmonoxid-Synthase (NOS)-positiven Subpopulationen ergab, dass die Mehrzahl der kompressions- und dehnungssensitiven Neurone in beiden untersuchten Colonabschnitten ChAT exprimierte, wobei diese ChAT-positiven Nervenzellen vorwiegend mit dem Neurotransmitter SP (ChAT / SP) colokalisiert vorlagen. Eine Immunoreaktivität für NOS zeigten MEN hauptsächlich im Colon transversum.
Es lässt sich schlussfolgern, dass porcine submuköse Neurone in der Lage sind, unterschiedliche mechanische Stimuli (Kompression und Dehnung) wahrzunehmen und mit neuronaler Aktivität auf diese zu reagieren. Außerdem konnte gezeigt werden, dass der exzitatorische Neurotransmitter ChAT bei der Erregungsübertragung der MEN dominiert. Die in dieser Studie gewonnenen Ergebnisse helfen, die komplexen Vorgänge der Mechanotransduktion porciner submuköser Neurone besser zu verstehen. Um die Frage nach der Eignung des porcinen Darms als Modell für den Menschen zu beantworten, ist es in Zukunft sinnvoll, weitere Daten, beispielsweise aus Ussing-Kammer Versuchen, zu erheben und mit zu berücksichtigen.

Characterization of mechanosensitive neurons in the submucosal plexus of the porcine colon
The enteric nervous system (ENS) contains two ganglionated plexuses and is located directly inside the wall of the gastrointestinal tract. Hence, its neurons are permanently exposed to the activity of circular and longitudinal muscles. In addition to neurons of the myenteric plexus (MP), which is located between the two intestinal muscle layers, it is known that neurons of the submucosal plexus (SMP) located in the submucosa are directly activated by mechanical stimuli. Data concerning the mechanosensitivity of porcine submucosal neurons are lacking so far, although the pig is discussed and used as a relevant model for human studies based on structural and physiological similarities of the gastrointestinal tract.
The aim of the present study was to gain basic knowledge about the neurons of the SMP in the porcine colon, with a special focus on their mechanosensitivity. First, a protocol was established that allows the visualization of neuronal signals in vital specimens of the proximal and transverse colon. The imaging technique used was ultrafast neuroimaging combined with the voltage sensitive dye Di-8-ANEPPS. Thus, all neurons of a ganglion could be examined at single cell level simultaneously. As the ganglia of the SMP of both examined intestinal localizations could already be identified in transmitted light of the light microscope by their size, morphology and surface structure, a preliminary staining of the neurons was not necessary. The viability of selected ganglia was proven by electrical or chemical stimulation of the neurons (using a point stimulation electrode and the nicotinic acetylcholine receptor agonist nicotine, respectively). Both methods successfully activated neurons in the examined colon sections. An essential part of the present work was mechanical stimulation. This included compression by intraganglionic volume injections as well as stretching of submucosal neurons. A specially developed and self-constructed stretching tool allowed the bidirectional stretch of entire ganglia in opposite directions for the first time.

Both stimuli reproducibly resulted in electrical discharges in a subpopulation of enteric nerve cells, whereby neither age- nor sex-related effects could be detected.
Immunohistochemical examination of mechanosensitive enteric neurons (MEN) with respect to their choline acetyltransferase (ChAT)-, substance P (SP)- and nitric oxide synthase (NOS)-positive subpopulations revealed that the majority of compression- and stretch-sensitive neurons expressed ChAT. These ChAT-positive neurons were predominantly colocalized with the neurotransmitter SP (ChAT / SP). NOS-positive MEN were mainly detected in the transverse colon.
It can be concluded that porcine submucosal neurons are able to perceive different mechanical stimuli (compression and stretch) and react to them with neuronal activity.
Furthermore, the excitatory neurotransmitter ChAT dominates in the excitation transmission of MEN. The results obtained in this study help to understand the complex processes of mechanotransduction of porcine submucosal neurons. In order to answer the question, whether the pig is an appropriate model for humans, it is necessary to conduct further experiments (for example Ussing chamber) to gain more data on functional properties of porcine colonic functions.
