Aufgabe 1 11 Punkte
Die in der Schweiz für Kosmetika zugelassenen UV-absorbierenden Inhaltsstoffe sind in einer Verordnung namentlich aufgeführt. Andere Substanzen sind verboten. Entwickeln Sie eine Methode, um die acht wichtigsten UV-Filter zu quantifizieren. Sie sollen auch nachzuweisen, dass keine unerlaubten Substanzen vorhanden sind.
Ein Hersteller analytischer Geräte hat eine HPLC-Methode entwickelt, die es erlaubt, die Substanzen zu trennen:

Die Substanzen auf Seite 3 wurden in gleicher Konzentration in Eluent A + Eluent B 1:1 gelöst und in den Chromatographen eingespritzt. Dabei wurde folgendes Chromatogramm erhalten.

Other compounds are not permitted. Develop a method to quantify the eight most important UV filters. You are also supposed to verify that no forbidden compounds are present.
A manufacturer of analytical instruments has developed an HPLC method to separate the compounds.

d) Kosmetika sind oft komplexe Gemische. Wie würden Sie das Ausgangsmaterial aufarbeiten, um schliesslich eine Flüssigkeit in den Chromatographen einspritzen zu können? (2 Punkte)
e) Was könnte die saubere Quantifizierung eines UV-Absorbers vereiteln? Wie würden Sie dem Problem begegnen? Könnte allenfalls der Detektor helfen? (3 Punkte)

Sie haben mit grossem Aufwand eine Substanz (unten) synthetisiert, von der Sie 10 mg reinstes Material für weitere Versuche benötigen. Sie haben 500 mg Ausgangsmaterial mit einer Reinheit von ca. 50%. Sie wollen die Reinigung möglichst kostengünstig durchführen.

a) Which kind of chromatography does this system represent? What is the separation criterion? (2 points)
b) What do you think of this separation system? Do you give this scheme any chance to solve your problem? What are the advantages and disadvantages, provided the scheme works at all? (3 points)
c) Independent of the usefulness of the scheme, critically consider the gradient. As no
experiment has been performed so far, you would certainly have to optimize it. What would you adjust first? (2 points)

Auf den folgenden Seiten finden Sie das IR-, Massen-, 1H-NMR- und 13C-NMR-Spektrum der Verbindung D5. Sie hat die relative Molmasse Mr = 161.

2a) Erklären Sie die Entstehung der Signale bei m/z 30 und 103 im Massenspektrum. Wenden Sie alle Fragmentierungsregeln an, die die Signale stützen, allenfalls mehrfach. (5 Punkte) b) Ordnen Sie die Signale im 1H-NMR-Spektrum den entsprechenden Protonen von D5 zu.
Die Signale bei 1.6 ppm und 1.5 ppm können auch vertauscht zugeordnet werden. Eine Begründung ist nicht notwendig. Beachten Sie auch den Hinweis in Aufgabe c). (4 Punkte) c) Erklären Sie das Aufspaltungsmuster der Signale bei 3.8 und 3.4 ppm im 1H-NMR-
Spektrum. Hinweis: Beachten Sie die Geometrie des Moleküls (unten). Die Teilspektren sind weitgehend 1. Ordnung mit einem klar erkennbaren Teil höherer Ordnung. Alle Signale können verstanden werden. (3 Punkte)

d) Ob die Protonen der NH2-Gruppe mit den benachbarten Protonen der CH2-Gruppe koppeln, hängt von den Aufnahmebedingungen ab. In CDCl3 ist die Kopplung oft nicht zu sehen, weil Verunreinigungen im Lösungsmittel einen schnellen chemischen Austausch der NH2-Protonen katalysieren. Ist die Kopplung sichtbar? Begründen Sie. (2 Punkte)
e) Die Aufspaltungsmuster der Signale bei 1.6 und 1.5 ppm im 1H-NMR-Spektrum sind nicht ohne Weiteres zu verstehen. Warum nicht? (1 Punkt)
f) Im Infrarot-Spektrum erscheint eine Bande für die N–H-Streckschwingungen der NH2- Gruppe bei 3370 cm–1. Weniger klar ersichtlich sind andere Banden zwischen 3400 und 2600 cm–1, (teilweise verdeckt durch die Banden um 3000 cm–1) die auch zur NH2-Gruppe gehören. Spekulieren Sie, warum die letzteren Banden einen so grossen Bereich einnehmen.
Hinweis: Eine grosse Verdünnung der Probe ändert wenig am Spektrum. (2 Punkte) g) Erklären Sie die sehr hohe chemische Verschiebung der CH-Gruppe im 13C-NMR-
Spektrum. (1 Punkt)

On the following pages you find the IR-, mass, 1H-NMR and 13C-NMR spectrum of compound D5. It has the relative molecular weight Mr = 161.

2a) Explain the formation of the signals at m/z 30 und 103 in the mass spectrum. Apply all fragmentation rules supporting the signals, more than once if necessary. (5 points)
b) Assign the signals in the 1H-NMR spectrum to the protons in D5. The signals at 1.6 and 1.5 ppm may be assigned interchanged. A justification is not required. Note the hint provided in task c). (2 points)
c) Explain the coupling pattern of the signals at 3.8 and 3.4 ppm in the 1H-NMR spectrum.
Hint: Consider the geometry of the molecure (below). The partial spectra are essentially first order with a clearly recognizable part of higher order. All signals can be understood. (3 points)

d) Whether the protons of the NH2 group couple with the neighboring protons of the CH2 group depends on the acquisition conditions. The coupling is often not present in CDCl3 because impurities in the solvent catalyze a fast chemical exchange of the NH2 protons. Is the coupling recognizable? Justify. (2 points)
e) The splitting pattern of the signals at 1.6 and 1.5 ppm in the 1H-NMR spectrum are not easily understandable. Why not? (1 point)
f) A band for the N–H streching vibrations of the NH2 group appears in the infrared spectrum at 3370 cm–1. Less obvious are other bands between 3400 and 2600 cm–1, (partially covered by the bands around 3000 cm–1) also belonging to the NH2 group. Speculate why these bands cover such a wide range. Hint: a large dilution of the sample does not change the spectrum substantially. (2 points)
g) Explain the large chemical shift of the CH group in the 13C-NMR spectrum. (1 point)
