Aufgabe 1 14 Punkte
Auf den folgenden Seiten finden Sie das IR-, Massen-, 1H-NMR- und 13C-NMR-Spektrum der Verbindung D10. Die Verbindung hat die relative Molmasse Mr = 264.

Hinweis zum 1H-NMR-Spektrum: Die Vergrösserungen auf Seite 4 sind in der Horizontalen gleich skaliert, so dass Linienabstände direkt verglichen werden können. In der Vertikalen sind die Skalen hingegen willkürlich.
Hinweis zum Massenspektrum: Signale unterhalb m/z 30 wurden nicht aufgenommen.
a) Das IR-Spektrum wurde bei relativ kleiner Konzentration aufgenommen. Spekulieren Sie, wie sich das Spektrum verändern könnte, wenn die Konzentration erhöht wird, abgesehen von den allseits stärkeren Banden. Erwarten Sie überhaupt eine wesentliche Veränderung?

b) Bei ca. 3000 cm–1 im IR-Spektrum findet man eine Vielfalt von Banden der C–H-Streck- schwingungen, was aufgrund der Vielzahl von CH-Umgebungen nicht verwundert. Sonst findet man keine breiten Banden. Insbesondere die Signale bei 1600 und 1520 cm–1 der aromatischen Gerüstschwingungen sind sehr scharf. Erklären Sie den Grund. (1 Punkt) c) Jemand vermutet, dass es sich bei der Bande bei 3420 cm–1 im IR-Spektrum um einen
Oberton der Carbonyl-Streckschwingung handelt. Was ist Ihre Meinung? (1 Punkt) d) Ob die Kopplung des Protons 5 im 1H-NMR-Spektrum zu seinen Nachbarn 4 sichtbar ist,
hängt auch von den Aufnahmebedingungen ab. Identifizieren Sie die Signale der Protonen 4 und 5 im Spektrum und analysieren Sie die Aufspaltungsmuster. Ist die Kopplung sichtbar?
Hinweis: Die Banden von anilin-artigen N–H-Protonen zeigen oft eine Verbreiterung, die auf die Kopplung mit dem 14N-Kern zurückzuführen ist. Das erschwert die Analyse. Der Effekt hat aber keinerlei Auswirkungen auf die Signale der Kopplungspartner. (3 Punkte) e) Ordnen Sie die restlichen Signale im 1H-NMR-Spektrum den Protonen zu. Eine
Begründung ist nicht nötig. Die Banden der H-Atome 7–10 können gemeinsam den Signalen zugeordnet werden. (4 Punkte)
f) Erklären Sie den Peak bei m/z 58 im Massenspektrum. Wenden Sie die Fragmentierungs- regeln, die das Signal stützen, möglichst oft an. (3 Punkte)

Remark on the 1H-NMR spectrum: The expansions on page 4 are scaled equally, so line distances can be compared immediately. The vertical scale is arbitrary.
Remark on the mass spectrum: Signals below m/z 30 have not been recorded.
a) The IR spectrum was recorded at relatively low concentration. Speculate on how the spectrum could change if the concentration was increased, besides all bands becoming stronger. Do you expect a substantial change at all? (2 points)
b) There is a variety of bands from the C–H streching vibrations at about 3000 cm–1 in the IR spectrum. Considering the multitude of CH moieties this is not astonishing. Elsewhere there are no broad bands. In particular the signals at 1600 und 1520 cm–1 of the aromatic skeletal vibrations are very sharp. Explain the reason. (1 point)
c) Somebody suspects the band at 3420 cm–1 in the IR spectrum to originate from an overtone of the carbonyl stretching vibration. What's your opinion? (1 point)
d) Whether or not the coupling between the proton 5 in the 1H-NMR spectrum and its neighbors 4 is visible also depends on the recordung conditions. Identify the signals of the protons 4 and 5 in the spectrum and analyze their splitting patterns. Is the coupling visible?
Hint: The bands of aniline-type N–H-protons often show broadening due to the coupling with the 14N nucleus. This makes analysis more difficult. However, the phenomenon has no effect on the signals of the coupling partner. (3 points)
e) Assign the remaining signals in the 1H-NMR spectrum to the protons. A justification is not necessary. The bands of the H atoms 7–10 may be assigned collectively. (4 points)
f) Explain the peak at m/z 58 in the mass spectrum. Apply the fragmentation rules supporting the signal as often as possible. (3 points)

Für die Verbindung D10 werden die alternativen Strukturen 1 und 2 vorgeschlagen. Finden Sie für jede Alternative mindestens zwei spektroskopische Argumente, die gegen sie sprechen. Ihnen stehen nur die Spektren und die jeweils falsche Struktur zur Verfügung. Vergleiche mit der richtigen Struktur sind unzulässig.
(1 Punkt für jedes Argument, maximal 2 Punkte pro Alternative.)

Aufgabe 3 5 Punkte
Die Papierchromatographie wurde Ende des 2. Weltkrieges eingeführt und erlebte eine stürmische Entwicklung während 30 Jahren (Nobelpreis 1952). Das verwendete Filterpapier besteht im Wesentlichen aus Zellulose. Bei der klassischen Papierchromatographie ist die stationäre Phase aber nicht Zellulose, sondern ein dünner Film von Wasser, der sich um die Fasern der Zellulose bildet. Als Laufmittel dienen wenig bis mittelpolare organische
Lösungsmittel. Typisches Beispiel: Essigsäureethylester : Essigsäure : Wasser (3 : 1 : 1).
a) Um welche Chromatographie handelt es sich bei der klassischen Papierchromatographie?
Wie funktioniert sie? Welcher Mechanismus führt zu einer Trennung von Analyten? Was ist das Trennkriterium? (3 Punkte)
b) Man kann als Laufmittel auch reines Wasser verwenden. Es kommt zu einer Trennung für gewisse Analyten, aber sie ist meist schlecht. Um welche Art Chromatographie handelt es sich? Welches Phänomen führt zur Trennung? (2 Punkte)

Paper chromatography was introduced at the end of World War II and experienced a rapid development in the following 30 years (Nobel prize 1952). The employed filter paper consists principly of cellulose. In classic paper chromatography the stationary phase is not cellulose, but a thin film of water formed around the fibers of the cellulose. Low to medium polar organic solvents are used as mobile phase. Typical example: Acetic acid ethyl ester : acetic acid : water (3 : 1 : 1).
a) Which kind of chromatography is classic paper chromatography? How does it work? What mechanism leads to separation of analytes? What is the separation criterion? (3 points) b) It is possible to use pure water as mobile phase. There is a separation between certain
analytes, but it is mostly poor. What kind of chromatography is this? What phenomenon leads to separation? (2 points)

Enantiomere (Bild und Spiegelbild) werden im Allgemeinen von chromatographischen Systemen nicht getrennt.
a) Wie muss grundsätzlich ein chromatographisches System aufgebaut sein, damit Enantiomere getrennt werden? (1 Punkt)
b) Welche chromatographischen Verfahren zur Trennung von Enantiomeren gibt es?
Beschreiben Sie mindestens zwei. Wie funktionieren Sie? (2 Punkte)
c) Kann man prinzipiell ein ionenchromatographisches System zur Trennung von
enantiomeren Anionen bauen? Wenn ja, wie könnte das funktionieren? Wenn nein, warum geht das nicht? Spekulieren Sie. (2 Punkte)

a) In principal, how can a chromatographic system work for separating enantiomers? (1 point) b) What chromatographic methods exist to separate enantiomers? Describe at least two. How
do they work? (2 points)
c) Is it possible, in principle, to build an ion chromatographic system to separate enantiomeric anions? If yes, how could it work? If no, why can't it work? Speculate. (2 points)
Aufgabe 5 8 Punkte
Viele Kunststoffe benötigen Zusatzstoffe, um geeignete mechanische Eigenschaften aufzuwei- sen. Polyvinylchlorid (PVC) ist einer der meist verwendeten Kunststoffe. Ohne Zusätze ist er hart und spröde. Um Flexibilität zu erreichen, können Weichmacher zugesetzt werden. Es besteht aber die Gefahr, dass die Substanzen den Kunststoff verlassen und in die Umgebung gelangen.
Es besteht der dringende Verdacht, dass eine Klasse Weichmacher, Phthalsäureester, siehe Seite 10, hormonelle Wirkungen entfalten, wenn sie in den Menschen gelangen. PVC-Folien werden zum Verpacken von Lebensmitteln verwendet. Sie müssen mit Weichmachern versetzt sein, um die richtigen mechanischen Eigenschaften zu besitzen. Es gelten daher strenge Vorschriften über die maximale Konzentration der Weichmacher in den verpackten Lebensmitteln.
a) Entwerfen Sie ein Verfahren, um die Weichmacher auf S. 10 in PVC zu quantifizieren.
Aufarbeitung, Trennung, Detektion, Quantifizierung. Erwartete Konzentration: mehrere Prozent. Hinweis: PVC löst sich gut in Tetrahydrofuran. (3 Punkte)
b) Entwerfen Sie ein Trennverfahren, um die Weichmacher in verpackten Fleischprodukten zu quantifizieren. Aufarbeitung, Trennung, Detektion, Quantifizierung. Erwartete Konzentra- tion: einige μg/kg. Wo erwarten Sie die grössten Schwierigkeiten? Glauben Sie, dass Sie das Problem mit den in der Vorlesung besprochenen Methoden überhaupt vollständig lösen können? Begründen Sie. (4 Punkte)
c) Einige der Substanzen auf S. 10 haben ein oder mehrere chirale Zentren. Inwiefern hat das einen Einfluss auf das von Ihnen vorgeschlagene Analysensystem? (1 Punkt)
Problem 5 8 points
Many plastics require additives to obtain suitable mechanical properties. Polyvinyl chloride (PVC) is among the most used plastics. Without additives it is hard and brittle. In order to gain flexibility, plasticizers can be added. However, there is a danger of these substances leaching from the plastic, thereby getting into the environment.
There is a strong suspision that a class of plasticizers, phthalic acid esters, see page 10, are endocrinic disruptors, if they get into the human body. PVC films are used to wrap food
products. They have to contain plasticizers to obtain the proper mechanical properties. There are rigid regulations about the maximum concentration of plasticizers in wrapped food.
a) Develop a (single) method to quantify the plasticizers on page 10 in PVC. Pre-processing, separation, detection, quantification. Expected concentration: several percent. Hint: PVC is well soluble in tetrahydrofuran. (3 points)
b) Develop a method to quantify the plasticizers in wrapped meat products. Pre-processing, separation, detection, quantification. Expected concentration: a few μg/kg. Where do you expect the most difficulties? Do you believe you can completely solve the problem with the methodology introduced in the lecture? Justify. (4 points)
c) Some of the substances on page 10 have one or more chiral centers. How far does this influence the analytic method you propose? (1 point)
