Ein Ziel dieser Dissertation war es, zwei Gruppen paternaler Halbgeschwisterkühe, die alternative, vom Vater ererbte BTA18 (43-48 Mb und 53-59 Mb) Haplotypen trugen, hinsichtlich Unterschieden in klinischen Parametern zu evaluieren. Zu den wichtigsten Ergebnissen dieses Abschnitts zählt, dass Kühe, die von ihrem Vater den mit einem niedrigerem und somit vorteilhaften SCS assoziierten Haplotyp (Q) geerbt hatten, eine geringere Inzidenz für postpartale Krankheiten wie Mastitis und Metritis zeigten als ihre paternalen Halbschwestern, die von ihrem Vater den mit einem höherem und somit nachteilhaften SCS assoziierten Haplotyp (q) geerbt hatten. Des Weiteren ließen die signifikant höheren Serumkonzentrationen an BHB und signifikant niedrigeren Plasmakonzentrationen an IGF-I der q-Kühe vermuten, dass sie sich schlechter an die metabolischen Herausforderungen der Laktation adaptieren konnten als ihre Q-Halbschwestern. Außerdem zeigten die Q-Kühe eine bessere Milchleistung (ECM) und Futteraufnahme als die q-Kühe. Für die Übertragbarkeit der wissenschaftlichen Ergebnisse in die Zuchtpraxis sind diese Studienergebnisse günstig. Es eröffnet Perspektiven Milchkühe zu züchten, die zwar weniger anfällig für postpartale Krankheiten wie z.B. Mastitis sind, aber eine mindestens ähnliche Milchleistung wie das Populationsmittel aufweisen.
Das eigentliche Selektionsziel des Q/q-Modells basiert auf einer unterschiedlichen somatischen Zellzahl der divergenten Haplotypen. Für dieses Merkmal liefert die Studie auf den ersten Blick konträr zueinanderstehende Ergebnisse. In der Frühlaktation fielen die q-Kühe vermehrt mit Eutervierteln mit extrem niedrigen Zellzahlen (SCC) auf. In der Literatur wird kontrovers diskutiert, ob eine Mindestanzahl an lokalen Immunzellen zur Erregerabwehr notwendig ist (RAINARD et al. 2018). Zur Mitte und zum Ende der Laktation hingegen war der SCS im Gesamtgemelk der q-Kühe signifikant höher als bei ihren Q-Halbschwestern, wie die Zuordnung der Färsen vor ihrer ersten Laktation in die Q- bzw. q-Gruppe prognostiziert hatte. Der Migration von Zellen in die Milch liegen komplexe physiologische und immunologische Mechanismen zugrunde, so dass eine einfache Bewertung des SCC/SCS nicht möglich ist. Die Ergebnisse dieser Studie zeigen, dass in die Evaluierung des SCC und SCS als Merkmal für Mastitisresistenz sowohl der Zeitpunkt der Bestimmung des SCC bzw. SCS miteinfließen als auch die individuellen Unterschiede auf Viertel- und Euterebene berücksichtigt werden sollten.
Abschließend lässt sich zusammenfassen, dass die Q-Kühe sowohl ihre immunologische als auch ihre metabolische Kompetenz betreffend bessere Fähigkeiten zeigten als ihre paternalen q-Halbschwestern. Inwieweit Immunsystem und Stoffwechsel abhängig vom Genotyp dabei miteinander agieren, ist bisher nur unzureichend erforscht. Aber es gibt in der Literatur bereits Hinweise, dass die Leber
unter der Doppelbelastung durch die Pathogenabwehr und Energieproduktion nicht mehr alle ihre metabolischen Aufgaben erfüllen kann (MOYES et al. 2016).
Außerdem wurden in dieser Dissertation die Effekte einer intramammären Pathogeninfektion mit Mastitiserregern (S. aureus und E. coli) auf das Lebertranskriptom laktierender Milchkühe analysiert. Die intramammäre S. aureus-Infektion resultierte in einer massiven Reaktion des Lebertranskriptom, obwohl die S. aureus-Mastitis in der Literatur als lokale Infektion i.d.R. ohne systemische Effekte auf den Wirt beschrieben wird (SCHUKKEN et al. 2011; JENSEN et al. 2013; HE et al. 2016; SONG et al. 2016). Dabei standen offensichtlich Adaptationen des Aktin-Zytoskeletts im Vordergrund, aber auch wichtige Netzwerke des Immunsystems waren von der intramammären S. aureus-Infektion beeinflusst.
Die Untersuchung des Lebertranskriptoms nach intramammärer E. coli-Infektion kann in zwei Kernergebnisse zusammengefasst werden: 1.) Das Komplementsystem war im Vergleich zur Kontrollgruppe nach E. coli-Infektion dramatisch runterreguliert, während die Akute Phase Reaktion aktiviert wurde, um den eindringenden Erreger zu bekämpfen. Dahinter könne ein bisher unbekannter Mechanismus von E. coli stecken, das Immunsystem des Wirts entscheidend zu schwächen und so das eigene Überleben zu sichern. Die Inaktivierung des Komplementsystems könnte aber auch einem wirtseigenen Regulationsmechanismus zugrunde liegen, der zum Ziel hat, die massive Aktivierung der Akute Phase Reaktion auszugleichen. 2.) Neben den Auswirkungen auf immunologischer Ebene waren auch Veränderungen in metabolischen Netzwerken (Fett- und Energiestoffwechsel) zu beobachten. Der Aspekt einer erhöhten metabolischen Herausforderung des Tieres durch pathogenspezifische Anpassungen des hepatischen Transkriptoms könnte ein neuer Ansatz für die Behandlung und Prävention der Krankheit sein.
Abschließend sollte diese Dissertation Leberfunktionen identifizieren, die nach intramammärer Pathogeninfektion (S. aureus oder E. coli) zwischen paternalen Halbgeschwisterkühen mit alternativen, vom Vater ererbten BTA18 (43-48 Mb und 53-59 Mb) Haplotypen unterschiedlich ablaufen. In der E. coli-Gruppe überlagerte die pathogen- und tierspezifische Reaktion auf die intramammäre Infektion sämtliche Unterschiede zwischen den alternativen Haplotyp-Gruppen. Für die mit S. aureus behandelte Gruppe konnten allerdings Unterschiede im hepatischen Transkriptom sowohl in Bezug auf immunphysiologische wie auch metabolische Funktionen zwischen den Q- und q-Kühe festgestellt werden. Q-Kühe zeigten dabei ein stärker aktiviertes Immunsystem (gekennzeichnet durch die höhere Expression bzw.
Anreicherung der entsprechenden Gene bzw. Netzwerke) als ihre paternalen q-Halbschwestern, was auf eine höhere immunlogische Kompetenz, die besser eindringende Krankheitserreger abwehren kann, der Q-Kühe hindeutet.
6. ZUSAMMENFASSUNG

Charakterisierung des Einflusses divergenter Prädisposition für Eutererkrankungen sowohl auf klinische Parameter im Verlauf der Laktation als auch für die Reaktion der Leber auf eine Infektion der Milchdrüse
Ziel dieser Dissertation war es, Unterschiede in klinischen Parametern und im Lebertranskriptom zwischen zwei Kuhgruppen, die von ihrem Vater mit divergenten Empfänglichkeiten für Mastitis assoziierte Haplotypen auf Bos taurus Autosom (BTA) 18 (43-48 Megabasen, Mb und 53-59 Mb) ererbt hatten, zu untersuchen. Kühe, die von ihrem Vater den mit einem niedrigerem SCS (somatic cell score) assoziierten Haplotyp (Q) geerbt hatten, zeigten im Verlauf der Laktation tatsächlich einen signifikant niedrigeren SCS als ihre Halbschwestern und eine geringere Inzidenz für postpartale Krankheiten. Die Kühe hingegen, die von ihrem Vater den mit einem höherem SCS assoziierten Haplotyp (q) geerbt hatten, fielen in den ersten Wochen der Laktation durch vermehrte Euterviertel mit extrem niedrigen Zellzahlen auf.
Außerdem zeigten die q-Kühe signifikant höhere Serumkonzentrationen für BHB (β-Hydroxybutyrat) und signifikant niedrigere Plasmakonzentrationen für IGF-I (insulin-like growth factor-I) als ihre Q-Halbschwestern. In einem Infektionsversuch wurden definierte Euterviertel beider Haplotyp-Gruppen intramammär mit Staphylococcus aureus (S. aureus) oder Escherichia coli (E. coli) infiziert und Leberproben wurden für Trankriptomanalysen gewonnen. Die E. coli-Infektion und deren Varianz überlagerten die Effekte durch die divergenten Haplotypen. Die Q-Kühe reagierten auf die intramammäre S. aureus-Infektion mit einem stärker aktivierten Immunsystem in der Leber, gekennzeichnet durch die höhere Expression wichtiger Immungene (z.B. HMOX1, IGSF11, ICAM3) und die Anreicherung biologischer Netzwerke, die diese einbezogen. Q-Kühe schienen daher ein umfangreicheres Repertoire an Abwehrmechanismen gegen eindringende Pathogene abrufen zu können.

Characterization of the influence of divergent predisposition for udder diseases on clinical parameters during lactation as well as on liver response to mammary gland infection
The aim of this dissertation was to investigate differences in clinical performance and at liver transcriptome level between two groups of cows which had inherited from their father alternative haplotypes on Bos taurus Autosom (BTA) 18 (43-48 Megabases, Mb and 53-59 Mb) associated with divergent susceptibilities to mastitis.
Cows that had inherited from their father a haplotype associated with a lower SCS (somatic cell score; Q) indeed showed significantly lower SCS than their half-sibs during lactation and fewer postpartal diseases. Cows, who had inherited from their father a haplotype associated with a higher SCS (q), were noticed in the first weeks of lactation to have more udder quarters with extremely low cell counts. In addition, q cows showed significantly higher serum concentrations for BHB (β-hydroxybutyrate) and significantly lower plasma concentrations for IGF-I (insulin-like growth factor-I) than their Q half-sisters. In a challenge experiment, both haplotype groups were intramammary inoculated with Staphylococcus aureus (S. aureus) or Escherichia coli (E. coli) and liver samples were collected for subsequent transcriptome analysis. The E. coli pathogen challenge and its variance overlaid the effects of the divergent haplotypes. The Q cows reacted to the intramammary S. aureus challenge with a more strongly activated immune system in the liver, characterized by the higher expression of important immune genes (e.g., HMOX1, IGSF11, ICAM3) and the enrichment of biological networks involving them. Q-cows therefore seemed to have access to a more extensive repertoire of defense mechanisms against invading pathogens.
