5.1. Analytische Verfahren
5.1.2. Massenspektrometrie
Für die Spektrenaufnahme wurden die Massenspektrometer Bruker Esquire-LC und Finnigan MAT 8200 verwendet. Folgende Ionisierungsmethoden wurden verwendet:
-Elektronenstoß-Ionisation (EI) -Chemische Ionisation (CI) -Elektrospray-Ionisation (ESI)
Die m/z-Werte entsprechen den Maxima der Isotopenmuster. Verbindungen, welche einen closo- oder nido Carborancluster enthalten, sind gekennzeichnet mit *. Die Identifizierung der Isotopenmuster dieser Verbindungen in den Massenspektren erfolgte durch Vergleich mit berechneten Isotopenverteilungen des „Isoform“-Programmes.
5.1.3. Infrarotspektroskopie
Die Aufnahme der IR-Spektren erfolgte mit einem Bio-Rad FTS 155 Spektrometer.
Die Messung erfolgte mit einem KBr-Pressling. Die relativen Intensitäten wurden mit br (breit), s (stark), m (mittel) und w (schwach) bezeichnet.

Es werden 6 g (19,4 mmol) N-Acetylneuraminsäure (4) in 380 ml wasserfreiem Methanol gelöst, welches 1,48 ml (19,9 mmol) Trifluoressigsäure enthält. Das Reaktionsgemisch wird unter Stickstoff 40h bei TR gerührt bis in der DC (1-Propanol : Wasser; 7:3; v:v; Rf = 0.53) die Ausgangsverbindung nicht mehr nachweisbar ist.
Methanol und Trifluoressigsäure werden i.Vak. entfernt. Es werden 6,04 g (18,7 mmol) weißes, amorphes Produkt erhalten.

Es werden 6,02g (18,6 mmol) Methylester 34 in 120 ml frisch destilliertem Acetylchlorid bei 0°C gelöst, welches 60 ml Essigsäure enthält. Nachdem eine klare Lösung erhalten wurde, wird das Reaktionsgemisch weitere 36 h bei TR gerührt bis die DC (Ethylacetat; Rf {38} = 0.29) kein Edukt mehr zeigt. Acetylchlorid und Essigsäure werden i.Vak. entfernt. Dabei darf die Temperatur des Wasserbades 40°C nicht übersteigen. Zu dem Reaktionsprodukt wird mehrmals trockenes Toluol gegeben und i.Vak. verdampft. Es werden 9,65 g (18,1 mmol) einer weißen amorphen Substanz erhalten.
Ausbeute: 98%

Es werden 3,89 g (12,0 mmol) Methylester 34 in 72 ml Pyridin gelöst. Zu der Lösung werden 48 ml Essigsäureanhydrid gegeben und als Katalysator 120 mg 4-(Dimethylamino)pyridin [DMAP] zugesetzt. Pyridin und Essigsäureanhydrid werden i.Vak. bei 45-51°C entfernt. Das Rohprodukt in Form eines braunen Sirups wird in 78 ml Dichlormethan gelöst, dreimal mit jeweils 15 ml gesättigter Natriumhydrogencarbonatlösung und einmal mit 30 ml 1N Salzsäure sowie 45 ml gesättigter Natriumchloridlösung gewaschen. Es werden 4,58 g (8,58 mmol) weißes amorphes Produkt erhalten.

Zu einer Lösung von 9,25 g (18,1 mmol) Chloroglycosid (38) und 12,6 ml ( 13,2 g; 122,3 mmol) Benzylalkohol in 114 ml Dichlormethan werden bei TR 6,78g (27,7 mmol) frisch hergestelltes Silbersalicylat gegeben. Das Reaktionsgemisch wird im Dunkeln 5h bei TR gerührt. Dann wird der Niederschlag abfiltriert und das Filtrat mit eiskalter 5%iger wäßriger Natriumhydrogencarbonatlösung, mit 5%iger wäßriger Natriumthiosulfatlösung und mit Wasser gewaschen. Das Filtrat wird über Natriumsulfat getrocknet. Nach Entfernen des Lösungsmittels und eines Teils des Benzylalkohols i.Vak. wird ein dunkelbraunes Öl als Rohprodukt erhalten. Nach der Säulenchromatographie (Chloroform : Aceton; 5:1; v:v; Rf der DC: ) werden 6,83 g (11,7 mmol) reines Benzylglycosid (39b) als amorpher weißer Feststoff erhalten.

Es werden 3,50 g (6,02 mmol) Benzylglycosid (39b) in 70 ml trockenem Methanol gelöst. Es werden wenige Tropfen 1N Natriummethanolatlösung hinzugegegeben, bis der pH-Wert 9 erreicht wird. Danach wird das Reaktionsgemisch 20 h unter Stickstoff bei TR gerührt. Weil der pH-Wert auf pH 7 gefallen ist, wird weitere Natriummethanolatlösung hinzugegeben bis pH 9 wieder erreicht wird. Die Lösung wird weitere 4 h gerührt und dann mit Amberlite IR 120 (H+-Form) neutralisiert. Das Lösungsmittel wird i.Vak. entfernt. Es bleiben 2,11 g ( 5,10 mmol ) weißes amorphes Produkt zurück.

Zu 2,45 g (5,93 mmol) Benzylglycosid (40b) wird zweimal je 30 ml trockenes Pyridin gegeben und jeweils i.Vak. verdampft, um Wasserspuren zu entfernen. Danach wird das Edukt in 55 ml Pyridin gelöst, die Lösung auf 0°C gekühlt und 1,13g (5,93 mmol) p-Toluolsulfonsäurechlorid in kleinen Portionen hinzugegeben. Das Lösungsmittel wird i.Vak. entfernt. Zum Rückstand werden 8 ml Eiswasser und 40 ml Ethylacetat hinzugegeben. Die organische Phase wird separiert, während die wäßrige Phase noch zweimal mit je 25 ml extrahiert wird. Die vereinigten organischen Phasen werden eingeengt. Es werden 2,285 g (4,03 mmol) einer braunen amorphen Substanz erhalten.

Nach der Chromatographie und dem Entfernen des Laufmittels i.Vak.
erhält man 786 mg (1,79 mmol) Reinprodukt.

In einem 100 ml Rundkolben werden 600 mg (1,41 mmol) Azid (43b) in 25 ml Wasser gelöst. Als Hydrierkatalysator wird 62 mg Palladium-(II)-oxid verwendet. Das Reaktionsgefäß wird mehrfach mit Wasserstoffgas gespült, ehe es unter leichtem Überdruck – der Rundkolben wird mit einen Ballon verbunden- mit dem H2-Gas befüllt wird. Das Gemisch wird 4 h bei TR schnell gerührt und dann filtriert. Das Filtrat wird lyophilisiert und ergibt 504 mg (1,27 mmol) weißes amorphes Produkt. Ob Kristallwasser enthalten ist, wurde nicht bestimmt, ist aber laut Lit. zu erwarten.

In 1,5 ml trockenem DMF werden 100 mg (251 µmol) 9 NH2-Neu5Ac2αBn (44b) gelöst. Bei 0°C werden zunächst 76 µl (55,5 mg; 548 µmol) frisch destilliertes Triethylamin (55), danach 47,3 mg (230 µmol) o-Carboranylcarbonsäurechlorid (45), gelöst in 0,9 ml trockenem DMF, hinzugetropft. Das Reaktionsgemisch wird 40 min bei 0°C unter N2 gerührt, ehe die Erwärmung auf TR zugelassen wird. Während der

Reaktion tritt ein weißer Niederschlag auf. Nachdem der Ansatz 1h bei TR gerührt wurde, werden das Lösungsmittel und die Base bei TR im Ölpumpenvakuum entfernt.
Das Reaktionsprodukt wird zunächst in 10 ml Acetonitril digeriert. Der unlösliche Rückstand besteht größtenteils aus dem Neuraminsäureedukt und Triethylammoniumchlorid. Die in Acetonitril lösliche Fraktion wird nach Entfernen des Acetonitrils i.V. in einem zweiten Schritt mit 5 ml Diethylether gewaschen. In Lösung geht hierbei ausschließlich das Carboranedukt. Nach der Säulenchromatographie (Ethylacetat : Methanol; 1:1; v:v; Rf = 0.70) wird das Anion des Produktes erhalten, in dem das closo-Zwischenprodukt (46b) nur noch mittels MS (ESI) nachgewiesen werden kann. Das Triethylammonium-Anion wird während der Chromatographie vollständig durch Protonen ausgetauscht. Man erhält 13 mg Produkt, welches zum Natriumsalz umgesetzt wird.

Nach Zugabe von 2,88 g (11,8 mmol) Silbersalicylat wird das Reaktionsgemisch 5 h im Dunkeln bei TR. gerührt. In einem Gemisch aus 10 ml Tetrahydrofuran und 500 µl Wasser werden 93 mg (146 µmol) silyliertes 2-N-Carboranoylamido-glucopyranosid 51 gelöst. Nach der Zugabe von 8,5 mg (44,7 µmol) Toluolsulfonsäure (Monohydrat) wird das Reaktionsgemisch 2 h bei TR gerührt. Die Reaktionskontrolle erfolgt mittels Dünnschicht-chromatographie (1 : 1; Hexan : Ethylacetat). Nach 2 h 45 min wird es auf 60°C erhitzt, nach 3 h 30 min werden weitere 8 mg (421 µmol) Toluolsulfonsäure zur Lösung hinzugegeben. 5 h 30 min nach dem Start der Reaktion wird das Lösungsmittel i.Vak. bei zunächst 40°C, dann bei 60°C entfernt. Es bleibt 43 mg (123 µmol) hellbraunes Produkt zurück, welches mit 16,5 mg Toluolsulfonsäure verunreinigt ist. Eine Aufreinigung des Reaktionsproduktes erfolgt nicht.

Es werden 66 mg (166 µmol) 9-NH2-Neu5Ac2αBn 44b in 900 µl Pyridin gelöst, bevor das Silylisierungsmittel zugegeben wird. Das Reagenz enthält 160 µl (644 µmol) N,O-Bis(trimethylsilyl)acetamid, 41 µl (322 µmol) Chlortrimethylsilan und 137 µl (644 µmol) Hexamethyldisilazan. Mit einem CaCl2 –Trockenrohr wird Luftfeuchtigkeit ausgeschlossen. Das Reaktionsgemisch wird 1 h bei TR, danach 1h bei 40°C gerührt.
Die Silylisierungsmittel werden durch Destillation im Wasserstrahlvakuum entfernt.
Danach wird der größte Teil des Trimethylsilylacetamid durch Sublimation im Ölpumpenvakuum entfernt. Der zurückbleibende Feststoff wird mittels ESI-Massenspektrometrie und 1H-NMR-Spektroskopie [CD3CN und CDCl3] untersucht.
Massenspektometrisch kann nur das eingesetzte Edukt 44b nachgewiesen werden.
Der spektroskopische Nachweis von silylierten Benzylglycosiden gelang nicht.

In einem Dreihalskolben werden 10,2 g (70,1 mmol) ortho-Carboran (47) in 950 ml absoluten über Natrium getrocknetem Diethylether gelöst. Die Lösung wird auf -78°C herabgekühlt. Innerhalb von 20 min werden unter Rühren 30,7 ml einer 2,5 molaren Lösung von n-Butyllithium (76,8 mmol) in Hexan hinzugetropft. Bei gleicher Temperatur wird das Gemisch 20 min weiter gerührt. Ca. 20 bis 25 g Trockeneis wird in kleine Stücke gebrochen und zum Reaktionsgemisch gegeben. Nach 2 weiteren Stunden Rühren bei -78°C lässt man den Kolbeninhalt auf TR erwärmen. Das Lösungsmittel wird i.Vak. bei TR entfernt. Danach wird 300 ml Wasser zum Reaktionsprodukt gegeben und noch vorhandenes Carboranedukt durch Extraktion mit zweimal 150 ml Hexan entfernt. Nachdem die wässrige Phase mit 3 N Salzsäure angesäuert wurde, wird sie viermal mit je 150 ml Hexan extrahiert. Die vereinte organische Phase wird über Magnesiumsulfat getrocknet. Nach dem Entfernen des Lösungsmittes i.Vak. werden 13,2 g (70,1 mmol) einer weißen amorphen Substanz erhalten.
Ausbeute: quantitativ (Lit.: 95%)

Es werden 8,00 g (42,5 mmol) Carbonsäure (48) in 36 ml trockenem Toluol unter Stickstoff gelöst. 9,23 g (44,3 mmol) Phosphorpentachlorid werden innerhalb von 1h zur Lösung hinzugegeben. Nachdem das Reaktionsgemisch 30 min gerührt wurde, wird das Lösungsmittel und Phosphoroxychlorid bei 110°C und Normaldruck abdestilliert. Die Aufreinigung erfolgt mittels Destillation im Ölpumpenvakuum (79°C bei 0,5 mbar). Das Produkt erstarrt bei Normaldruck zu einer weißen amorphen Substanz, welche charakteristisch nach Säurechlorid riecht.
