Abstract

The leukocyte-heme enzyme myeloperoxidase (MPO) has emerged as an important mediator of inflammation. Being a peroxidase enzyme, it generates hypohalous acids, which are not only involved in killing pathogens but are also capable of modifying proteins and affecting cellular function. Independently of its catalytic function, MPO also possesses unique physical properties. Upon release, MPO is found deposited along the endothelium where it mediates neutrophil recruitment and activation, solely dependent on its cationic surface charge. The vascular endothelium is lined with sugar and protein molecules called the endothelial glycocalyx (EG). The EG plays a crucial role in maintaining vessel function and physiology. Given the importance of the EG and MPO’s deposition in the endothelium, we aimed to investigate MPO’s extra-catalytic properties further and its effect on EG integrity. A murine cremaster model was established to study EG integrity in terms of thickness using intravital microscopy. In C57BL/6J (WT) mice, EG thickness reduced significantly with both externally administered and neutrophil-secreted MPO. A similar effect was also observed with catalytically inactive MPO (Q91T) and cationic polypeptide, poly-lysine, suggesting the effect to be dependent on ionic interactions and independent of MPO’s enzymatic property. This was also observed via in vivo staining of the glycocalyx with Alcian Blue that clearly showed a decrease in the anionic charge of the EG upon MPO/Q91T/poly- lysine binding. The in vivo reduction of the EG was further investigated where removal of bound MPO led to restoration in EG thickness, reconfirming the physical nature of the phenomenon. However, the modulation of the EG did induce shedding of some of EG components, namely syndecan-1 (Sdc1) proteoglycan, which was not directly MPO-mediated but rather dependent on MPO-induced neutrophil recruitment. Lastly, in vitro studies with Chinese hamster ovary cells revealed heparan sulfate glycosaminoglycan (GAG) as MPO’s binding partner, thus responsible for its ionic interaction with the EG. In summary, MPO leads to a collapse of the EG structure via physical binding with heparan sulfate GAG, thereby causing Sdc1 shedding in a neutrophil-dependent manner. These findings extend MPO’s chemotactic role based on its cationic surface charge that not only induces inflammation but also critically influences vascular integrity.

Zusammenfassung

Das leukozytäre Häm-Enzym Myeloperoxidase (MPO), welches die Generierung von reaktiven Spezies katalysiert, wurde in der Vergangenheit als ein wichtiger Mediator von Entzündungsprozessen identifiziert. Als Peroxidase katalysiert es vor allem die Bildung von hypochloriger Säure, die nicht nur an der Abtötung von Pathogenen beteiligt ist, sondern auch in der Lage ist, Proteine, Lipide und DNA zu modifizieren und die Zellfunktion zu beeinflussen. Unabhängig von seiner katalytischen Funktion besitzt MPO auch einzigartige physikalische Eigenschaften. Nach Freisetzung aus Leukozyten wird MPO entlang des Endothels abgelagert, wo es Neutrophilenrekrutierung und -aktivierung vermittelt, allein abhängig von seiner kationischen Oberflächenladung. Das Gefäßendothel ist mit Zucker- und Proteinmolekülen ausgekleidet, die endotheliale Glykokalyx (EG) genannt werden. Die EG spielt eine entscheidende Rolle bei der Aufrechterhaltung der Gefäßfunktion und -physiologie. Angesichts der Bedeutung der Interaktion von EG und MPO im Gefäß wollten wir die extrakatalytischen Eigenschaften von MPO und seine Wirkung auf die EG-Integrität weiter untersuchen. Ein Maus-Cremaster-Modell wurde entwickelt, um mit Intravitalmikroskopie die EG-Integrität, d.h. die Dicke der EG, zu untersuchen. In C57BL/6J (WT)- Mäusen reduzierte sich die EG-Dicke signifikant sowohl mit extern verabreichtem als auch mit aus Neutrophilen sekretiertem MPO. Ein ähnlicher Effekt wurde auch mit katalytisch inaktivem MPO (Q91T) und kationischem Polypeptid, Polylysin, beobachtet, was darauf hindeutet, dass der Effekt von ionischen Wechselwirkungen abhängt und unabhängig von der enzymatischen Eigenschaft von MPO ist. Dies wurde durch in-vivo-Färbung der Glykokalyx mit Alcianblau bestätigt, die eine deutliche Abnahme der anionischen Ladung der EG nach MPO-Bindung, sowie nach Bindung von Q91T und von Polylysin zeigte. Der Einfluss auf die EG in-vivo wurde weiter untersucht, wobei die Entfernung von gebundenem MPO durch Heparin zu einer Wiederherstellung der EG-Dicke führte, was die physikalische Natur des Phänomens weiter bestätigte. Die Modulation der EG induzierte jedoch die Abspaltung einiger EG- Komponenten, nämlich von Syndecan-1 (Sdc1) Proteoglycan, das nicht direkt MPO-vermittelt, sondern eher von MPO-induzierter Neutrophilenrekrutierung abhängig war. Schließlich zeigten in-vitro-Studien mit Ovarialzellen des chinesischen Hamsters (CHO Zellen) Heparansulfat-Glycosaminoglycan (GAG) als

Bindungspartner von MPO, das somit für seine ionische Wechselwirkung mit der EG verantwortlich ist.

Zusammengefasst führt MPO zu einem Kollaps der EG-Struktur durch physikalische Interaktion mit Heparansulfat-GAG, wodurch die Sdc1-Freisetzung auf von Neutrophilen abhängige Weise bewirkt wird.

Diese Befunde erweitern die Erkenntnisse zu extrakatalytischen Eigenschaften von MPO, die nicht nur Entzündungen induzieren, sondern auch die vaskuläre Integrität entscheidend beeinflussen.
