Abstract

Abstract

Overexpression and increased activity of the small Rho GTPase Rac1 has been linked to squamous cell carcinoma of the epidermis and mucosa in humans.

Targeted deletion of Rac1 or inhibition of Rac1 activity in epidermal keratinocytes reduced skin tumour formation in a chemical skin carcinogenesis mouse model.

However, a potential role of Rac1 in UV-light induced skin carcinogenesis has not been investigated so far, solar UV radiation being an important carcinogen to the skin.

To investigate this, we deleted Rac1 or modulated its activity in the epidermis of mice using Cre/lox mediated targeted deletion of the rac1 gene and/or expression of dominant negative or constitutively active mutants of Rac1. In mice with additional transgenic expression of the complete early region of Human papilloma virus type-8 (HPV-8) in epidermal keratinocytes we observed strong reduction of spontaneous or UV- light induced skin papilloma formation upon Rac1 deletion/ inhibition and a strong increase in spontaneous skin papilloma frequency upon constitutive activation of Rac1. Surprisingly, however, in spite of reduced skin papilloma formation, most of the mice with epidermal Rac1 deficiency developed squamous cell carcinomas of the skin upon long-term UV- irradiation. This shows a protective function of Rac1 in UV-light induced skin carcinogenesis.

Abstract

Short-term UV-irradiation experiments revealed increased UV-light induced apoptosis of Rac1 deficient epidermal keratinocytes along with an increased inflammatory response in skin with Rac1 deficient epidermis. Restoration of Rac1 activity in Rac1 deficient epidermal keratinocytes rescues increased UV-light induced apoptosis only in vitro but not in vivo, suggesting contribution of additional non-cell autonomous mechanisms to keratinocyte apoptosis in vivo.

Further investigations revealed that the observed increased UV-light induced apoptosis is DNA damage dependent and that Rac1 deficient keratinocytes show an attenuated DNA damage response upon UV-irradiation.

Taken together, our data provide first direct evidence for a protective role of Rac1 in UV-light induced skin carcinogenesis and keratinocyte apoptosis probably through regulating mechanisms of the DNA damage response and repair pathways.

Zusammenfassung

Plattenepithelkarzinome der Haut und Schleimhaut des Menschen zeigen Überexpression bzw. Überaktivierung der kleinen Rho GTPase Rac1. Gezielte Deletion von Rac1 oder die Hemmung von Rac1-Aktivität in epidermalen Keratinozyten reduzierten die Papillomentstehung in einem Modell für chemische Karzinogenese der Haut. Ein wichtiges Karzinogen für die Haut ist ultraviolettes (UV) Licht; bisher war unbekannt, ob Rac1 auch eine Funktion in der UV-Licht- induzierten Karzinogenese der Haut hat.

Um dies zu untersuchen, haben wir Rac1 gezielt in der Epidermis von Mäusen mit Hilfe des Cre/lox-Systems deletiert oder seine Aktivität durch transgene Expression dominant negativer oder konstitutiv aktiver Mutanten moduliert. In Mäusen, die gleichzeitig die frühe Region des Genoms des Humanen Papillomvirus Typ 8 exprimierten, führte die Deletion/Hemmung von Rac1 zu einer Reduktion der Entstehung von Papillomen der Epidermis, während die konstitutive Aktivierung von Rac1 zur spontanen Entstehung von Papillomen führte.

Obwohl der Verlust/die Hemmung von Rac1 in der Epidermis zur Reduktion der Papillom-Entstehung führten, kam es in Langzeitexperimenten mit UV- Bestrahlung beim grössten Teil der Mäuse mit Epidermis-spezifischer Deletion von Rac1 zur Entstehung von Plattenepithelkarzinomen der Haut. Dies zeigt eine protektive Rolle von Rac1 in UV-Licht induzierter Karzinogenese der Haut.

Abstract

Experimente mit einer einmaligen UV-Bestrahlungsdosis zeigten, dass Rac1- defiziente epidermale Keratinozyten eine verstärkte Apoptose nach UV- Bestrahlung aufweisen. In vivo ging dies mit einer verstärkten Entzündungsreaktion der Haut in Mäusen mit Epidermis-spezifischer Deletion von Rac1 einher. Wiederherstellung der Aktivität von Rac1 in Rac1-defizienten Keratinozyten führte zu einer Normalisierung der UV-induzierten Apoptose in vitro, jedoch nicht in vivo. Dies legt einen Beitrag nicht-zell-autonomer Mechanismen zur verstärkten UV-Licht induzierten Apoptose von Rac1- defizienten Keratinozyten nahe. Weitere Untersuchungen zeigten, dass die beobachtete verstärkte Apoptose abhängig von UV-induzierten DNS-Schäden ist und dass Rac1-defiziente Keratinozyten eine abgeschwächte Reparaturantwort auf DNS-Schäden nach UV-Bestrahlung zeigen.

Zusammenfassend zeigen unsere Daten erstmalig eine protektive Rolle von Rac1 in der UV-Licht-induzierten Karzinogenese und Apoptose der Haut, wahrscheinlich durch Regulation von Mechanismen der DNS-Reparatur nach UV-Bestrahlung.
