4. DISKUSSION

4.5. Ausblick

Die wichtigste, noch zu klärende Frage ist weiterhin die Rolle von CusF in der cus-vermittelten Kupferresistenz. Um dies zu untersuchen, ist es notwendig die fehlenden Liganden bei der Bindung des Kupfers an CusF zu identifizieren. Bei CusF handelt es sich bei anscheinend um ein neuartiges Kupferzentrum, für das nach Kristallisation des Proteins die an der Kupferbindung beteiligten Aminosäurereste identifiziert werden könnten. Auch wäre zu untersuchen, welche Aminosäurereste von CusF für die Interaktion mit dem Transportkomplex wichtig sind. Desweiteren ist zu klären, ob es sich bei CusF tatsächlich um ein frei im Periplasma bewegliches Kupfer-Chaperon handelt, oder ob CusF Bestandteil des CusCBA-Efflux-Komplexes ist.

5. ZUSAMMENFASSUNG

Kupfer ist auf Grund seiner Redoxeigenschaften eines der wichtigsten Spurenelemente, doch ist auf dieses Redoxverhalten auch seine Toxizität zurückzuführen. In Escherichia coli wurden bisher drei chromosomal kodierte Systeme identifiziert, die an der Kupferhomöostase beteiligt sind: Die P-Typ-ATPase CopA transportiert Cu(I) aus dem Cytoplasma in den periplasmatischen Raum. Die Multicopper-Oxidase CueO schützt periplasmatische Enzyme vor Kupfer-induzierten Schäden. Das Cus-System wird durch zwei Operone, cusRS und cusCFBA, kodiert, die in Gegenrichtung transkribiert werden. Das cusRS-Operon kodiert für ein Zwei-Komponenten-Regulationssystem, cusCFBA für die Proteine eines CBA-Efflux-Komplexes und für CusF. Ziel dieser Arbeit war die Untersuchung der cusCFBA-vermittelten Kupferresistenz, die in E. coli nur in Abwesenheit von CueO bestimmbar ist.

1. Durch Expression von cusA bzw. cusCFBA in trans konnten die E. coli Stämme GR10(∆cueO ∆cusCFBA::cm) bzw. GR15(∆cueO ∆cusA::cm) partiell komplementiert werden. Ohne CusB und CusC war CusA nicht in der Lage, im E. coli Stamm GR10 Resistenz zu vermitteln. Die Deletion von cusC, cusF oder cusB in E. coli GR1(∆cueO::cm) hatte eine Verminderung der Kupferresistenz zur Folge. Dabei führte eine Deletion von cusB zu gleich starkem Resistenzverlust, wie die Deletion von cusA. Demgegenüber zeigte ein Stamm mit Deletion von cusC eine geringe Restresistenz. Den geringsten Einfluss hatte die Deletion von cusF, doch konnte auch hier eindeutig die Beteiligung an der Kupferresistenz gezeigt werden.

2. Als RND-Protein besitzt CusA 12 Transmembran-Durchgänge und zwei große periplasmatische Domänen. In der zweiten periplasmatischen Domäne konnten Methioninreste als potentielle Liganden des Kupfers identifiziert werden. Diese Reste sind auch in anderen RND-Proteinen konserviert, die möglicherweise am Transport monovalenter Kationen beteiligt sind. Der Austausch der Methioninreste M573, M623

und M672 führte zum Verlust der Kupferresistenz und sind daher essentiell. Der Austausch von M812 zu Isoleucin führte zu einer Verringerung der Kupfertoleranz. Die Mutation anderer ebenfalls konservierter Methioninreste der zweiten periplasmatischen Domäne von CusA hatte keinen Einfluss auf die Funktion des Proteins.

3. Mutationen konservierter geladener Aminosäurereste in der Transmembran-Helix IV von CusA zeigten, dass die negativen Ladungen essentiell für die Funktion des Proteins sind. Demgegenüber hatte die Einführung einer zusätzlichen Ladung eine Verminderung der cus-vermittelten Kupferresistenz zur Folge.

4. CusF ist im Periplasma lokalisiert. Das Protein wurde nativ gereinigt. Die leader–

Sequenz wurde in vivo nach Aminosäure Q21 abgespalten, so dass ein prozessiertes Protein im Periplasma vorlag. Es wurden polyklonale Antikörper gegen CusF gewonnen, mit denen CusF im periplasmatischen Extrakt von E. coli nachgewiesen wurde. Die Expression von CusF erfolgte Kupfer-abhängig. Gereinigtes CusF-Protein band in vitro ein mol Kupfer pro mol Protein, war violett gefärbt und zeigte im UV/visible Spektrum ein Absorbtions-Maximum bei ca. 510 nm.

5. Ergebnisse des Yeast-Two-Hybrid-Systems deuten auf eine Interaktion von CusF mit CusB und CusC. Die Deletionen der Gene des cusCFBA-Operons führten zu einer erhöhten Cus-Expression vom Promotor cusCp, wobei keine Unterschiede zwischen den einzelnen Mutantenstämmen auftraten. Somit ist für CusF eine Rolle bei der Regulation des Promotors cusCp unwahrscheinlich.

6. Durch Expression von CusF in trans war es möglich, einen cusF-Deletionsstamm [E.

coli Stamm EC933(∆cueO ∆cusF)] partiell zu komplementieren. Mutation der konservierten Methioninreste M69 und M71 in CusF verminderte die Fähigkeit zur Kupferbindung auf 0.2 mol Kupfer pro mol Protein und die Komplementationsfähigkeit des Mutanten-CusF.

7. Mutation der Histidinreste von CusF zu Glutamin hatte keinen Einfluss auf die vermittelte Kupferresistenz. Nach Austausch der Histidinreste H57 und H58 zu Argininresten wurde das Protein nicht mehr prozessiert. Eine Fünffach-Histidin-Arginin-Mutante wurde im Cytoplasma akkumuliert. Auch die Mutation weiterer Aminosäurereste (D48N und F73Y) als mögliche Liganden des Kupfers, hatte keinen Einfluss auf die Funktion des Proteins.
